Replacing motherboard bios update help

Hello,
I'm replacing my broken motherboard, asus Maximus viii hero alpha, with the asus z170 deluxe.

I was wondering what I should do regarding updating the bios.
Should I unplug the drives that have my OS on so it doesn't affect it. And do the updating like that?
Also I was wondering about the whole windows may not activate thinking it's a new PC thing? What to do D:
First build please go easy on me :3

Motherboard has a USB BIOS Flashback Button.

Download and read motherboard manual from Asus. Instructions on renaming BIOS file are in there.
