Psu killed my motherboard

I built a new rig two days ago and it was working until booting windows, then it turned off and would no longer boot or post. Can I test the psu with a multimeter when it is not hooked up to anything?
 
Solution
You can do a very rudimentary test with a multimeter but, unfortunately it will not tell anything about how the PSU will act under load. It will however let you know if the PSU will function and if voltages are out of spec (+/- 5% for positive rails, +/- 10% for negative rails)

on a side note, if everything turns on and I get no boot led on motherboard, do you think it's the motherboard or CPU that broke, I have no other rigs to test anything on. And have already tried every possible way to reset mobo with no success, does not even recognize a video signal

When I contacted Asus they said the ram wasn't compatible but I had no problem with the ram. Just was not on their vendor list
