Just got a new SSD and was wondering if I have to format the D drive which is a partition of the main hard disk. I got the SSD as a boot drive. I really would like to keep all my data.

OK...so here is how to proceed.
1. Reduce the current used space in your C partition to below 90GB.
2. Clone over all partitions on the current HDD, except the D partition.

These steps should probably do it:

Verify the actual used space on the current drive is significantly below the size of the new SSD
Download and install Macrium Reflect (or Samsung Data Migration, if a Samsung SSD)
Power off
Disconnect ALL drives except the...

Well...what do you want the end state to be?
The OS and everything on the new SSD?
Or the SSD as just a secondary drive?

What size is the new SSD?
How much total used space is on your current C drive?
Desktop or laptop?

I'd like the boot drive the be the SSD and I'd like to keep my D drive which is also on the same disk as the current C drive.

My current C drive has a total of 149GB on it and my SSD has a total of 120GB but I'm only using 110GB on the current C drive.

OK...so here is how to proceed.

Power off
Disconnect ALL drives except the current C and the new SSD
Power up
Run the Macrium Reflect (or Samsung Data Migration)
Select ALL the partitions on the existing C drive, except the current D partition
Click the 'Clone' button
Wait until it is done
When it finishes, power off
Disconnect ALL drives except for the new SSD
Swap the SATA cables around so that the new drive is connected to the same SATA port as the old drive
Power up, and verify the BIOS boot order
If good, continue the power up

It should boot from the new drive, just like the old drive.
Maybe reboot a time or two, just to make sure.

If it works, and it should, all is good.

Later, reconnect the old drive and wipe as necessary.
Delete the original boot partitions, here:

Physically disconnect the drives or just disconnect in Disk Utility.

Physically disconnect.
Just open the side panel and take out the SATA cable.

But my C:\ drive is on the same hard disk as my D:\ drive.

So no problem.
Just do the steps above, being sure to not select the D partition in the process.
And after it is done, the first thing you do is to disconnect the current HDD before you power up the first time with the SSD.

So I'm in Macrium Reflect. What do I do in the program. It says create a backup on the screen I'm on.

Also one more thing. After a few boots from the SSD I should be able to just plug in the old Hard Drive and have all the Data on D?
