Asrock X-99 Extreme 3 - Help Adding M.2

My build is as follows:
i7-5820
AsRock X-99 Extreme 3 - latest bios
32 GB Ram
Transcend 512GB SSD (boot drive)
Samsung 960 EVO M.2 500GB (trying to set up as a boot drive)
Win 10 - latest

I installed the drive per the manual instructions, making sure nothing was attached to the SATA port 3_2 and setting that port to an SSD. I boot up. load the driver, run the cloning software and everything seems fine. I boot up again after removing the transcend drive and suddenly the M.2 drive has disappeared. I am totally lost here, after hours of tinkering it seems to come and go... Never able to get the PC to boot from the M.2 drive. Any ideas or links to others would help. The drive is so new and I am not that savvy with BIOS settings...

OK, got it back to a regular disk drive (not boot), so I am at least back in business. I tried creating a Windows 10 ISO on a flash drive for a fresh install, but it could not find the samsung driver to install. Is there some trick to that? Should I copy the driver directories onto the flash drive? Sorry, really over my head here...
