I am thinking of getting a samsung 960 evo 250GB m.2 ssd and a gtx 1070 graphics card. I would have both on a Gigabyte Z170N gaming 5 mini itx motherboard.

I have read some things about not being able to have both on the same motherboard. I did not fully understand what they meant. Will the motherboard be able to support the m.2 and the graphics card?

Thanks in advance.

You can have both the GPU and the M.2 drive populated in their respective slots. The only downside will be that you're going to loose out on the 2 SATA ports that are located at the top side of the board, i.e
maxresdefault.jpg

Those located beside the chipset heatsink or rather SATA ports 4 and 5.
