Should I have a PCI-E and a SATA and Which is Better?

For a consumer I haven't yet seen the need for PCIe based drives. I generally tell people to avoid them because many systems won't boot from a PCIe drive. SSD and the NVMe drives are fast enough for anything we do.

My setup is a 128GB SSD for my OS and most programs, a 256GB SSD for my steam folder, and a couple of large spinning 5900RPM drives for mass storage. My Steam drive is filling up so at some point I'll get a 1TB drive for my steam folder, and move the 256GB drive over for my OS. My OS drive is an older drive anyways so I should do this sooner rather than later.

NVMe drives are faster than SATA drives but are generally also more expensive.

For boot, you'll notice it. For gaming, the effects will be negligible. For other programs, it depends. Storage speed is only noticeable when there is prolonged of read or write operations, like copying a large file.
