How do I know if my motherboard supports the graphic card I want to buy?
I have ASUS M5A78L/USB3 and I'd like to buy MSI Radeon R9 380.
My motherboard has a PCIe x16 2.0 and the graphic card is a PCIe x16 3.0, will that work? Is there something else I need to think about?

Yes it's 100% compatible , and no loss in performance using it in a 2.0 slot.
