I have the Asus P6T Deluxe V2 mobo with Corsair XMS3 6GB (3x2GB) DDR3 1600 MHz (tripple channel)
and I want to add a Corsair Vengeance 8GB DDR3-1600MHz (single channel). Will it work fine and do I have to do something to make them work together?

Solution
that is a weird mixture of ram modules, supposedly your mainboard will accept 6 modules of ram

https://www.asus.com/Motherboards/P6T_Deluxe/

the ram compatibility list is there

https://www.asus.com/Motherboards/P6T_Deluxe/HelpDesk_QVL/

it says 6 modules, each module of 4 gbs to have a maximum of 24 gbs of ram, i don't think it will 100% work, could work but perhaps will detect it as a 1 gb ram module or less

have you considered to move to a newer platform with support for bigger ram modules? ideally 16 gbs is enough for at least 3 more years, but your idea of perhaps 12 slow gbs, well, not sure if can work or if is worth to try it
