I currently have an SSD (Samsung 850 Evo) plugged into the M.2 slot and one optical drive plugged in to what appears to be the SATA6G_3 slot. I recently tried to plug in another drive via SATA but the computer wouldn't recognize it. Confused, I tried plugging the new drive into all of the other available SATA ports, one at a time, to no avail. I then unplugged my existing drive that I knew worked and plugged in the new drive into the exact same port and, bam, it works. However, when I go to plug in the old drive to any of the ports, nothing.

I'm not entirely sure what to make of it. According to my mobo manual the M.2 slot is only supposed to disable SATA Port 1, but only ONE of them is working!

Many motherbd do not have hot-plugging turned on or available for the sata ports. Try installing the drive with the power off. next boot up into the bios and see if it is detected there. If it is you can save and exit to Windows and if it doesn't prompt you to initialize the new drive, you can run diskmgmt.msc and initialize and format it there.
