Motherboard BIOS Update

I had a quick question about updating the BIOS of my motherboard. It is an ASUS Motherboard that supports USB Bios flashback and I wanted to know if I should build the PC then update the BIOS or should I do it before I start building. If you think I should do it before I start building, what should I place my motherboard and power supply on?

I need to update the BIOS to fix an issue. What im asking though is do I assemble the PC first or do it before with just the motherboard and powersupply.
 
Which issue is it, that you know for sure you need a BIOS update?

You can do like you want, it´s not necessary to install the CPU with this board, but you can and it is more save because you see if the BIOS update was successful. I would install all hardware components and then use ez flash to update it within the BIOS.

It's a compatibility thing. For my CPU to work with my mobo I need to update the BIOS. Apparently for my case, I can plug in a USB with the file on it and then press a button on my mobo and it will update. Do I still do it all assembled?

Oh, okay so it doesn't matter right? If I assemble it or if I don't its all the same right?
