No, it will take just the one slot.

I think what you want to ask is if the graphics card (PCIe 3.0) will be compatible with the PCIe 2.0 (x16) on that mobo. The answer to that is that yes as well - they will be compatible as the slots are physically the same, with the difference lying in bandwidth (which will cause negligible bottleneck).
