Will my Gigabyte B250M-D3H mobo support 1x4GB stick of RAM?

I was wondering whether or not a single stick of 4GB of RAM would work on my dual channel B250M-D3H mobo and if it would be better to buy a kit of 2 RAM sticks rather than buy one now and then buy the same stick later in the future.

Better a kit, but single should work also. With a kit the compatibility of the two modules is guaranteed by the producer.
You'd have to use DDR3L, not DD3.
