I'm running (5) 6TB drives with 128MB cache in Raid 5 using
x299 chipset Intel Rapid Storage Tech

Raid 5 volume data stripe is set to 128kb (default) but Windows 10 disk management partition only allows 64kb allocation units max.

Are these two different things (data stripes and allocation units)?
And how do these data groupings best help each other for video/audio storage?

I understand the concept of block size and search speed, I can't find an explanation of the relationship between stripe and unit size and how it effects alignment for latency.

Solution
I would VERY careful to use Mobo RAID, as you can see in this forum, there is a lot of issues when replacing the Mobo (upgrade or replaced). There are so much incompatible in RAID mode between chipset of Intel Mobo

If your data is important then consider HW raid Card
