I have a 2x4 GB sticks of RAM, I was thinking of adding another 8 gigs, and I'm wondering if I can add a single stick into my motherboard, making it 16 gigs in total.

RAM currently in my PC: Ballistix Sport 8GB Kit (4GBx2) DDR3 1600 MT/s (PC3-12800) CL9 @1.5V UDIMM 240-Pin Memory BLS2KIT4G3D1609DS1S00

Single stick of RAM I was thinking of adding into my PC:Ballistix Sport 8GB Single DDR3 1600 MT/s (PC3-12800) CL9 @1.5V UDIMM 240-Pin Memory BLS8G3D1609DS1S00

Motherboard: ASUS Micro ATX DDR3 2000 AMD AM3 Plus Motherboard M5A78L-M/USB3

My motherboard supports 32 GB of RAM, and everything seems identical in each RAM stick. Would it be compatible?

First off, there isn't any guarantee adding RAM will work. RAM is sold in kits for a reason. They have been tested to make sure they will work together. I and many others here know that it will sometimes not work and have fielded more than a few questions here about adding RAM and it won't work. 95% sure it will work. Sometimes it won't. It's best to buy a new kit. Sell your old RAM on craigslist or something.

Compatible? As long as they are identical to your own RAM. Speed(MHZ), Timing(CL9) and voltage(1.5V)

Lastly your RAM is dual channel. That means that when you install two modules they work in tandem with each other. That 1600MHz is their effective speed. If you only install one module it will work at half speed.

Your plan isn't an optimal solution but it should work but it won't work the way it was designed to.

So, if I were to add another 2x4GB of RAM I should be fine?

Should be? Yes. The first off up there still applies.
