Ram slots, does it matter?

So, I'm confused when it comes down to which slots I should place my ram-sticks in. As of now, my 2 ram-sticks are placed in a1 - a2. PC won't boot if I try to start it with the ram-sticks in a1 - b1 or a2 - b2.
Dunno how big of a deal it is, but some tips/help would be appreciated.

For this mobo, Asus recommends using first a2-b2 slots. If it does not work for you (and neither in a1-b1), that could mean the ram sticks you got do not work in dual channel mode together for some reason. Did you get them as single kit? If yes, that could mean they were not paired as they should be.
Depends on motherboard. Some are quite rigorous on which slots are to be used and will not work when ram sticks are put in 'wrong' slots. Others just don't care and will work no matter where you put ram. But usually each motherboard should work fine when using only one stick in whatever slot, so best way to check if your motherboard has all slots working is to test single stick in each slot and see if it works that way.

My motherboard is Asus strix b250f gaming and it runs fine with one stick, no matter the slot, as you mentioned. But whenever I try to use a1 and b1 or a2 and b2 it doesn't boot and just gives me the ram_led(I forgot the actual name) light. Is there any notable difference if it runs in a1 and b1 or is it just the same as running it with a1 and a2?

Ballistix 16gb (8gb x2) in a single package if thats what you mean
 
Did you reset CMOS after changing stick positions but before trying to boot with new configuration? If you didn't, you should try that with a2-b2 setup. If still no go, and sticks are still under warranty, I would RMA them. Because in a1-a2 setup they are not running in dual channel mode, which decreases performance.

Me running them in a2-b2* might be solve my problem which I've had since 09/08. I'll give it a shot and come back.
