so im using the
Asus - STRIX Z270-E GAMING ATX LGA1151 Motherboard

and im using a

Samsung - 960 EVO 250GB M.2-2280 Solid State Drive for os then gonna use a mechanical drive for storage. My question is the motherboard has to m.2 slots
1. M.2 Socket 3 Type M (2242-2280) Supports PCIe3.0x4 mode
And
2. M.2 Socket 3 Type M (2242-22110) Supports SATA & PCIe 3.0 x 4 modes

So which one do i use, to get the most speed.
Also which SATA spot will i connect my mechanical drive to on the motherboard?

Solution
For M.2 it does not matter which you use both are capable of PCIE 3.0 X4. I have the same board and I use my 500gb 960 evo in the top M.2 slot, though like I said it does not matter as far as im aware. After you install the drive make sure in the BIOS it is set to use x4 pcie lanes, mine defaulted to only using x2 lanes which will limit performance a bit.

According to the manual all the SATA ports are 6gbps so agin it does not matter which you use, I use ports 1 and 2.

You can use both. But normaly you loose some SATA ports.

If you use nr. 1 (closest to the CPU) you loose 1 SATA port, normaly SATA port 1
If you use nr. 2 you loose 2 SATA ports, normaly SATA ports 5 and 6

Speed is the same, just make sure its set x4 speed and not x2 speed in bios.

Now which SATA port to plug a regular HDD into all depends on which M.2 socket are in use and so which SATA port is then disabled (look in your motherboard instruction book)
