Can I use FreeDOS as a main OS?

I have a bunch of old software that runs from disks, including games, and would love to install freedos. Are there any negatives to this setup as opposed to ms-dos 6.22 can I install it from a USB stick?

FreeDOS is, basically, DOS - text-only OS. You'll end up with a minuscule partition for DOS, with nothing else.

As you probably know, DOS does not support USB, you have to explicitely configure CD-ROM drivers, network, sound, etc. Your old games might have problems with hardware which came twenty years after them, extremely fast CPUs in particular.

DOS emulators like DosBOX are known to work well with old games.
