If I combine 2 sticks in dual channel with 1 in single channel, will the third one only be used when memory usage is maxed out

RAM is kinda finicky. Mixing and Matching RAM does not always work out. Even if you are mixing RAM that is of the same model. This is why RAM is sold in kits. They are tested together to work. Running single channel on a dual channel board can also cause problems.

But that does not mean it wont work, you will just have to try it. But I would buy the RAM from a vendor with a good return policy.

I'm aware of most of that but I'm more concerned about the technical aspects of how memory allocation actually works
