Does this motherboard need to use the top PCIE slot?

I recently built a new PC. Everything turns on- the fans spin, the motherboard and GPU light up but I get no signal on the monitor. I have the Asus ROG Strix 1080ti and a B250H motherboard. It would be a pain to put it on the 1st PCIE slot, but is that why it isn’t working? Thanks!
 
Solution
Rule out a graphics card issue. Next move would be leaving single RAM module installed and testing each slot. Keep it simple when troubleshooting. Minimum needed components to POST. You'd also narrow down the possible faulty components this way.

Thanks for the fast response. I’m using an i7-6700k. I’m won’t be at my PC until Monday so I can’t do that but what would that do?
