You will only need the PCIExpress slot, if you buy a PCIExpress card, instead of an AGP. Right now, there isn't anything to gain really for PCIExpress over AGP, the cards just can't use the extra bandwidth.
Also currently PCIExpress, for the most part is on mainly on Intel boards. AMD should start supporting this very soon though.
PCIExpress is sort of the new upcoming thing, but won't be a must if you buy an AGP card.

The X800 seies is available in the new PCIexpress slot and there is also the AGP8x versions available. Either slot, you will not notice a performance difference, since the card does not exceed the thoroughput of either bus.

After doing some searhing, I concur with Mr5Oh, I couldn't find and AMD 64 based boards with PCIExpress slots...You may want to look into replace you card with a AGP version, and look at getting a <A HREF="http://www.msi.com.tw/program/products/mainboard/mbd/pro_mbd_detail.php?UID=607" target="_new">MSI K8N NEO2 Platinum</A> Motherboard

What says that: "Supports up to 3500+, 3800+ Athlon 64 FX 53, or higher CPU"
One of the boards you were looking at? Your 3000+ 64-bit chip is socket 754? You may have been looking at a scoket 939 motherboard. Make sure you are looking at the right motherboard for you chip.
