It makes a difference. DV is supposed to be lower field first, but some
capture programs do not label things correctly, and some clips get their fields
reversed due to glitches in the source material.
So.
Best idea is to set up your procedure and test it both ways.

Does this apply to PAL as well as NTSC? I thought PAL used upper field but I
could be wrong?

PAL was upper field in most analogue systems, but redefined to lower
field first for DV (caused me no end of confusion when I first got a
DV edit system, kept trying to force it to upper field :-)
