I have 2 HDD's with multiple partitions.

Disk 1: C (Primary), K, L, M, N & O
Disk 2: E, F, G, H & I

What happens to the drive letters if I remove Disk 2, I may be replacing it
with a much larger one?

I do not know if the letters may change by default when you remove one disk,
but whatever happens you can reset them later. Go to start-programs-Admin
tools-Computer management-Storage-disk management then right click any disk
in the bottom right segment of the window to get an option to change the
drive letter.

Hi,

The drive letters will not change on the primary disk unless you elect to
change them (unlike Win9x where it automatically, and sometimes
disastrously, relettered the volumes). Volumes created on the new drive will
be given the first alphabetically available letters unless you designate
otherwise.
