external hard disk as master?

Can a hard disk be a master if it's in an external enclosure that connects via usb2?

You can boot off any kind of USB device your BIOS supports booting from, check your BIOS boot order.

I was wondering something similar. I need to run data recovering on my Mac's hard disk, but can't use the disk both to recover to and from, I need a second disk. Could an external disk (Trekstor) do, and can this external disk be set as master? How would this be done?
