Problem with Minolta Scan Elite 5400 using Vuescan

I have just paid for Vuescan to use with the 5400 scanner. I have just tried
to scan some mounted slides (using the mounted slide holder that came with
the scanner) and they have all scanned out of focus. If I scan unmounted
film using the unmounted film strip holder (supplied with the scanner) the
same images are in focus. If I use the original Minolta software the images
are in focus using both holders. I can only assume that I have the Vuescan
software set incorrectly. Two questions:-
1. Will Vuescan work with the mounted slide holder.
2. If so, what settings should I use in the Vuescan software
Sorry if these are stupid questions but I cannot find the answer in the
manual, either that or I am just blind.
