Yes, the Vista device manager shows the DJ 600 and I have it my printer manager. It lists LPT1 as the port. I have a HP Slimline. I have just USB ports, so I have an adapter from Centronics connector to USB.

The plotter sees the data and prints shaded rectangles in various line weights. The physical cable connection is ok.

HP does not support the DJ600 under Vista as shown on their device matrix somewhere in their website.

I was wondering if I should connect the plotter as a RS232 device.

Your type of connector will not fix your problem. You need a driver that HP didn't create. The only choices you have, that I can see, is either use an O/S that HP supports for this plotter, or get a plotter the supports Vista.

This is a problem that many computer people discovered with the implementation of Vista. Sorry I can't offer you a better solution.
