SLI has to be enabled in the drivers, and without two cards you couldn't do it. Even if you could "enable" it with only one card, it'd be exactly the same as running a single card.
The old SLI boards had a physical bridge chip that you had to reverse in order to go from 16x1 to 8x2 for SLI. So if you did that your card would be running on 8x instead of 16x but even that probably wouldnt have made much difference.
