Yea, so I'm building my PC, mobo is Asus p5n32-e and at the top there're 2x 4pin connecters , but one of them has a cover on it. My power supply has two of the plugs but the one that should go into the second 4 pin seems to have different shapes than is required. Should I only have one plugged in or am I being stupid to the max?

Cheers

The cover should only be removed if you have a 8-pin EPS +12V power plug. Only one of your PSU's 4-pin connectors should be plugged in there. Leave the cover on.
