i have 320gb hdd and I'm going to install windows vista, and i want to know if i should leave it all in One partition or should i brake it in two?
I would make the primary where the OS goes 70gb and the rest as second partition, I've heard that if its smaller where the OS is, it can perform better since it has less stuff to look at.

does HDDS work better when partition?

Depends on what you use the drive for. If you only use the second for backups then yes windows will run better since it really only works with one partition. Putting commonly used programs on the second partition will make the drive bouce back and forth between the partitions slowing it down. Best thing to do is use a second hard drive for programs and one drive for the OS.

How often do you do performance check, like defragging, cache cleaning and registry cleaning? If not done regularly, then at some point it will slow your computer down.

Some of my computers have the following partition:

1 OS - NTFS
2 Temp Drive - FAT32
3 DATA NTFS

Or

1 OS - NTFS
2 Games system -NTFS
3 Temp Drive - Fat32
4 Work or Data Drive NTFS

Where temp drive have the following files:
Page Files
Temporary internet Files
Temporary Download Files
Temporary work files
Windows Temporary Files

While some might argue that this is not a good solution, but it works for me.

Like sturm said 2 hard drives are better than 1.
