If I put my video card in the 2nd PCIe x16 slot will it still function? I am wanting to add a PCIe 1x TV tuner card to my system, and the primary PCIe x16 slot with a video card in it blocks the 1x slots.

no
you can only boot from video on top or a pci

you might get it in windows to work but you would need another monitor to configure it
