I am running a patch cat5e from my router to a wall plate. I ran inside/outside cat5e from the plate to a second plate on the first floor (45') then, a patch from the plate to a second computer. The computer will not get a signal. Do I have to wire one of the plates as a crossover (3,6,1,4,5,2,7,8)?

The cable does not must be crossover.

No, you shouldn't need a crossover cable.

For the most part the only time you need a crossover cable is when you connect two pc's together without a switch/hub/router.

Router --> Wall Plate --> Inside Wall --> Wall Plate --> Computer should be a normal patch cable at all points.
