Does the chipset matter if you use a dedicated pci-e card?

This is really gonna sound incredibly stupid.
I understand that each series of chipsets offer increases over the previous, but if I just want to throw in a PCI-E card to playa game. is there that much of a difference in each chipset to make up for the $50.00+ each series cost over previous?
