Hi,
I currently have an ECS Mini ATX board an I am planning on upgrading to an ASUS full ATX board for the extra slots and also the effect that it has on appearance (No empty space)

They are both the same Socket etc so I am just going to swap ram and everything over.

One problem, Will this require a reinstall of Windows?

I would suggest a reformat and fresh install of the OS and new MB system drivers/utilities. Otherwise a repair install would be needed due to the old MB's system drivers including chipset, sound, LAN, etc. presence on the HD. I have used both methods and found repair installations to be less desirable and sometimes problematic down the road. Save all the important data on the drive to disk or external drive and do a clean install with the new MB installation is my suggestion.

BTW, a full ATX MB will not fit in a micro specificied case.
