I have a desktop with the following hard drives:

Western Digital Raptor WD1500ADFD 150GB 10000 RPM SATA 1.5Gb/s 3.5" which I use as a backup for photos and music files.

Seagate Barracuda 7200.11 ST3500320AS 500GB 7200 RPM SATA 3.0Gb/s 3.5" which has my Windows Vista OS and all my programs and documents.

I'm getting a Synology DS110j and would like to use the 500 GB disk with it. Is it possible prior to formatting the 500 GB disk to copy my Windows folder and program files folder from the 500 GB to the 150 GB, then use the 150GB as my master HDD in the desktop so I won't need to reinstall Windows Vista and all my programs?

You will need another drive 300GB or more. Move photo and music files from 150gb to the 300GB plus drive. Shrink the 500GB under 150GB and then create a backup image using windows backup image creator to the 300GB plus drive. Set the 150GB for boot and restore image.
