When I fire it up for the first time, how are the drives assigned C:, D: and F: on an Asus PT6 Deluxe V2?
Does Sata 1, 2, 3, 4, 5 and 6 have anything to do with it?
Same thing for the 2 DVD burners?
SATA is new to me. Last build was with IDE.

Windows assigns the drives their letters, which ever drive you install windows onto will be labeled the C drive and its recovery partition lands on D, im not sure how it decides which one of the drives gets labeled next.
I forget the system used to assign letters but it is all decided in windows. if you do not likt the designations you can change them in disk management with the exception of the C: drive which can not be changed
