My motherboard specs state " PCI Express x16 Slot (compatible with PCI Express x8 speed only" ) Does that mean whether I have one or two graphics cards in, the speed will always be x8? I also wanted to get a gts 250 and was wondering how much an affect x8 speed will have on it.

Unless you have the older PCI-E 1.0/1.1 you wont see any loss in performance when running two cards in x8 mode.
If your using a single card, the slot will run at the full x16 link.
P.S. Could you please link the motherboard in question?
Quite a few older SLI 'boards ran 1 card at X16 but dropped to X8 for two, but the point is moot; The GTS250 is not fast enough to suffer even a tiny loss of performance at X8 speed.
So buy it and enjoy, mate.
