Configuring a Lenovo D20.

12 RAM slots.
Dual Intel XenonW5580 Processors.
Quadro FX4800

Future memory upgrade aside - price aside -
pros / cons: more RAM sets smaller total RAM each caard or fewer cards with more RAM each card?
ie: 4x1GB=total thus 4 cards total 4GB or 1 card 4GB on it?

You need at least one memory module per CPU. Using larger modules allows for easier expansion. Since that system is triple channel, you should install 3 or 6 modules per CPU. At a minimum you should install 2 modules per CPU for dual channel.
