I just upgraded to an ati 4350 from integrated graphics.
When i start up the machine, the machine POSTs (1 beep) and shows the asus m3a78-em motherboard screen.
"Press DEL to run Setup, Press TAB to display BIOS POST Message" is written at the bottom of the screen, but hitting DEL or TAB doesn't do anything and I'm just stuck on this screen.

Should I try resetting the BIOS? The machine worked fine until I added the GPU so i'm guessing all the other parts are ok

normall y a bios would autodetect you have a gfx card and start using it .

For some reason your mb is confused , and the best way to sort it out will be to find the setting in bios that disables the onboard gfx and disable it .
Just plugging the VGA/dvi cable back in to the mb socket should get you to the bios , but if it doesnt you may have to remove the card , make the changes and then re-install

1) First, unplug *all* USB cables and reboot. Glitches in USB hubs sometimes cause this type of freeze. If the reboot is successful, plug in all USB cables and continue.

2) If that fails, remove the vid card and go back to integrated graphics. Does it boot now? If so, use the opportunity to ensure your BIOS is setup to use the vid card. Shut down, reinsert the vid card, and try again.

3) If that fails, clear CMOS, enter BIOS, make whatever changes you need and save them. try Integrated graphics first, then insert the vid card and try that.

This sequence will give you a good chance to understand what went wrong.
