I have 3 SATA drives; a DVD, a 640 GB hard drive, and a 1TB hard drive. I am not doing RAID.

My Corsair TX750 PSU (single rail) has 2 SATA power cables.

Does it matter which cable powers each drive? Can they be on 1 cable or should I put the HDs on 1 cable and the DVD on the other?

Use whatever gives the most convenient layout. If one cable will reach all all four drives conveniently, just tie the other out of the way.

Theres enough capacity on that psu rail to power dozens of hard drives as well as the dvd

yeah, the single rail is actually preferable to PSUs that split the 12v rail into multiples.
