i have a ASUS P5P43TD Pro LGA 775 Intel P43 ATX Intel Motherboard mother board with and want to put in

EVGA 01G-P3-1155-TR GeForce GTS 250 1GB 256-bit GDDR3 PCI Express 2.0 x16 HDCP Ready SLI Supported

Video Card graphics card in would that work?

Yup, it will work fine.

The difference between PCI-E 1.0 and 2.0 is that 2.0 offers twice as much bandwidth but the GTS250 is far from getting bottlenecked by a PCI-E 2.0 8x slot so it will work perfectly, there shouldnt be any bottleneck at all.

Nope, far more powerful cards than yours show no bottlenecking going between PCI-E 2.0 16x and 8x(equivalent to PCI-E 1.0 16x)

That graphics card will not even come close to using up all the bandwidth of a PCIe 1.0 slot anyway so no bottleneck there.

It's been shown that the HD5870 1GB (current top performer for a single GPU card) is not limited using an 8x instead of 16x PCIe v2.0 interface.

In the system you describe, potential bottlenecks are your CPU and possibly your RAM but definitely not the PCIe bus.
