Using 1gb RAM With/Without 256 mb RAM , which is right?(DDR)

Hello. I'm Just newbie in hardware

Which right and wrong?
1GB DDR X 256 MB DDR= ??
1GB DDR W/O 256 MB DDR= ??

Some one told me that i take of the 256 MB RAM

Personally, i'd take out the 256mb stick. Run sets of the same RAM, rather than different sizes.
