Will i be able to use my SSD and Hard drive at the same time ?

Hello i was planning on getting a crucial m4 64GB SATAII MLC SSD and a Seagate Barracuda 1TB 7200 RPM 32MB Cache SATA 6.0Gb/s 3.5 Internal Hard Drive, and i was wanted to boot up windows through my SSD and store games, media files and software on my hard drive, would i be able to run both at the same time ? my specs so far are

ASrock extreme3 gen3 LGA 1155 socket SATA 6.0Gb/s motherbard

intel i5 2500k lga 1155 socket

GTX 560ti 2GB graphics card

corsair enthusiast 750W power supply

8GB g.skill RAM

coolermaster hyper 212 plus

plextor dvd burner

storm enforcer case
