Can you use a 2.5" HDD in a destop sata 3 motherboard?

Do you need an adapter to do this, because the connections do not look the same.

If you see a bunch of pins on the drive, you need an adaptor. Startech makes good adaptors for almost anything.

If its a SATA 2.5" you can plug it right into SATA and SATA power on the desktop. Do it at work all the time.
