To use an SLI setup, do you have to have a SLI certified motherboard, or can you set it up as long as you have multiple PCI Express x16 slots?

You need a motherboard with SLI support, the motherboard needs to have a chipset that supports SLI and the appropriate licensing to run SLI, nVidia charges for the license for a board to run SLI so not all boards that have chipsets that could support it do support it.

An SLi certified board is the easiest way, unless you want to play about with hacked BIOS's and drivers.
