If you replaced your current card with something that performs similarly, then you will not notice any immediate changes.If, however, you upgraded to a high performing card compared to the previous one, then you will not only get visible improvements in windows display effects/animations(provided that Aero is turned on in Windows Vista/7- if you have them) but also the performance in graphics applications will increase.

For example,if you go from an integrated card to a high-end card(like going from Intel 4500 to a HD 5770), you get a great boost in 3D apps/games (mostly games)

It is also very important that in order to see any gains that you uninstall the previous GPU drivers properly, before you install the new GPU and its drivers. Also, if your CPU is not up to the task of pumping sufficient data to the GPU, you may not see the performance leap you expect.
