I want to buy a USB 3 PCI (express) card soon. For USB 3 to work, does it need to be connected to a SATA 6 gb/s port? Or will the PCI connection suffice- in other words simple insert the card and voila? I have a MSI P55 CD53 motherboard.

The PCIe slot is all it needs data-wise. For external power, if it supplies it, it may need a molex power connector fro the PSU.

Yes, though you may have to install drivers before installing the card its basically plug and play.
