Does it make any difference on ram with 1x8 vs 2x4 vs 4x2 ?

1x8 will run in single channel which will be slower, and 4x2 will put more stress on your memory controller

No because 4x2 is still dual channel unless it's on a x79 chipset. 1x8 is best for if you or going to upgrade, but as said is single channel and is pointless to get more ram if it is a gaming build. 2x4, no complaints there, still allows 2 extra slots to upgrade. 4x2, as said is more stress on the mem controller plus you have to get rid of ram to upgrade which means money lost.
