In the manual it mentions there are special ways for the characters to
advance from, say, mage to wizard, etc. How can I do this? Also, after
defeating the vampire for the star gem, if I exit the room with the gem on
the lower right door and follow it around, there is a stone block in the
floor. What do I do with this?

Midway through the game. You'll know it when you get there.
