If it's an Intel stock cooler, it has 4 black plastic "screws" on it. If you twist those 45 degrees counter clockwise, the cooler should become loose. If it's still stuck after twisting those thingies, beware and remove the heatsink with extreme caution, as it may rip the cpu out of it's socket.
 
Like Chriscornell and 13thmonkey said but first unplug it from the board. Then after you twist the 4 corner clips pull each in turn up while holding the middle down and then give the HSF (heatsink/fan) a quarter turn to help it break loose of the thermal paste and then you should be able to pull it straight out.

Before you put any more thermal paste on the processor clean all of the old paste off with some coffee filters and some rubbing alcohol. Use the coffee filters to remove as much of the old paste as possible and then I use a Qtip dipped in rubbing alcohol to get the rest of the paste. Then use another coffee filter to clean the processor of anything left before you put it back in the socket.
