Hello,
I have an ASUS M4A78_EM mobo that I am currently using the onboard card for video. Now I want to upgrade and get a separate graphic card. My question is that I Have a PCI Express 16 slot. Is a PCI Express version 2.0 card compatible?

AMD Athlon X2 250 (3.0GB), 4GB of RAM with 750GB HDD; these are current specs

Yea a 2.0 card should work.Just try to stay away from 2.1 cards because their known to have issues with 1.0 slots.
