If I buy a single 8gb stick and my mobo is dual channel will I lose memory? Is there a big difference if I use 2x4gb?

No you will not loose any except 2x4GB would be slightly faster but not noticeably.

you will be operating in single channel mode. won't cause you any problems, although i would recommend a 2x4 setup.
